Osimhen scored from the penalty spot to level the match before the break.
The Florence team's midfielder Giacomo Bonaventura scored to give Fiorentina a 2-1 lead in the 63rd minute.
Fiorentina cemented a 3-1 victory after Nicolas Gonzalez scored in stoppage time.
The winning side leapfrogged into fourth position above the Naples club.
Fiorentina have 17 points in eight matches. Napoli have 14 in the Italian league led by AC Milan, who have 21 points.
